NAD is a dinucleotide: two nucleotides joined through their phosphate groups, one carrying adenine and the other nicotinamide. The nicotinamide ring is the reactive part, accepting a hydride to give the reduced form, NADH, and releasing it on reoxidation. As a redox cofactor it is consumed and regenerated in glycolysis, the tricarboxylic acid cycle and oxidative phosphorylation, and the NAD+/NADH ratio is a standard measure of cellular redox state. In its non-redox role it is a substrate rather than a catalyst: sirtuins, poly(ADP-ribose) polymerases, CD38 and SARM1 cleave and consume NAD, linking it to ADP-ribosylation, chromatin studies, DNA damage models and calcium signalling through cyclic ADP-ribose. The container holds a freeze-dried solid, made by freezing and removing the ice as vapour under reduced pressure. The bonds at risk are not peptide bonds: the pyrophosphate bridge and the nicotinamide glycosidic bond are the labile ones.
Redox state is the other reason the dry form matters. In solution the two species interconvert, and their pH sensitivities are opposite, the oxidised described as unstable in alkali and the reduced in acid, so redox composition depends on storage as much as manufacture. The solid is hygroscopic. Storage practice is a matter for the receiving institution; no guidance is supplied.
