Modified GRF (1-29) is the first twenty-nine residues of human growth hormone releasing hormone, C-terminally amidated, with four substitutions: D-alanine at 2, glutamine at 8, alanine at 15, leucine at 27. Ipamorelin is unrelated: a five-residue peptide of aminoisobutyric acid, D-2-naphthylalanine and D-phenylalanine, residues no ribosome incorporates.
The confusion here is the trade name on the first component. CJC-1295 no DAC is not CJC-1295 with DAC, CAS 446262-90-4, which carries an extra maleimidopropionyl lysine and weighs about 3647.2. Ipamorelin is grouped with GHRP-2 and GHRP-6, separate sequences with their own CAS numbers.
The components are studied at two receptors. Work on GRF(1-29) analogues concerns the GHRH receptor of pituitary somatotrophs, a class B receptor described by cyclic AMP generation. Work on ipamorelin concerns GHS-R1a, the ghrelin receptor. Raun and colleagues characterised it in rat pituitary cells, anaesthetised rats and conscious swine in the European Journal of Endocrinology in 1998, reporting growth hormone release without the corticotropin and cortisol changes seen with GHRP-6 and GHRP-2.
Those are measurements by others in cells and animal models. Receptor activity in a model does not establish an effect in a person, and no statement is made about the two together.
